In short: In 1972, a Nobel laureate's team faced a wall: they could synthesize vitamin B12, but they couldn't purify the specific isomers needed for the final stages. The solution required a breakthrough in separation technology that would eventually help build a global leader in analytical chemistry.

The synthesis of vitamin B12 is one of the most complex feats in organic chemistry. In 1972, Helmut Hamberger, a chief post-doc for Nobel laureate Robert Burns Woodward at Harvard University, sought to complete the first synthesis of the vitamin. The primary obstacle was the need to purify positional isomers—molecules that are nearly identical in structure but differ in the placement of specific atoms. Without high-precision separation, the chemical process could not reach its final stages.

To overcome this, Hamberger turned to Jim Waters, the founder of Waters Associates. Working together, the team developed a separation method in just two days. They then spent five additional days securing larger columns to scale up the process, followed by three more days of preparation. This collaborative effort successfully isolated and purified 200 mg of the precursor compound, a pivotal moment for both biochemistry and the commercial trajectory of Waters. This success highlighted the necessity of high-pressure, high-accuracy pumping and chromatography. Waters capitalized on this expertise, introducing the M6000 pump in the 1970s, which won awards for its precision. Expanding rapidly, the firm dominated 40% of the liquid chromatography sector by 1978, outsizing its closest rival fivefold. Today, the company provides the analytical instruments—such as mass spectrometry and chromatography systems—that allow scientists to identify metabolites in human serum, such as those used in COVID-19 research to identify biomarkers for renal dysfunction.

The evolution of these tools has moved from manual lab processes to high-resolution platforms like multi-reflecting time-of-flight (MRT) mass spectrometry. With sub-ppm mass accuracy, these contemporary setups deliver a mass resolution of over 200,000 Full Width at Half Maximum. This level of detail allows researchers to resolve overlapping lipid species and identify clinically relevant metabolites, continuing the legacy of precision separation that began with the synthesis of vitamin B12.
