Smooth muscle contracts slowly, but lasts longer than skeletal muscle

In short: Smooth muscle contracts more slowly than skeletal muscle but maintains tension longer due to its unique phosphorylation process. This allows it to sustain contractions for extended periods, essential for functions like digestion and blood pressure regulation, without requiring constant energy input.

Smooth muscle and skeletal muscle share some similarities, such as the presence of actin and myosin filaments, but their mechanisms of contraction differ significantly. While skeletal muscle relies on a precise sequence of events involving calcium release from the sarcoplasmic reticulum and the troponin-tropomyosin complex, smooth muscle uses a different pathway. Calcium enters the cell through L-type voltage-gated channels, triggering the release of more calcium from the sarcoplasmic reticulum. This calcium then binds to calmodulin, which activates myosin light chain kinase (MLCK).

MLCK phosphorylates the regulatory light chain on myosin, causing a conformational change that increases ATPase activity. This allows myosin to bind to actin, initiating the cross-bridge cycle and resulting in contraction. However, unlike skeletal muscle, where calcium removal leads to immediate relaxation, smooth muscle requires the action of myosin light chain phosphatase (MLCP) to dephosphorylate myosin. This means that even after calcium levels drop, the muscle remains contracted until MLCP acts, making the process slower but more sustained.

This slower, longer-lasting contraction is crucial for the functions of internal organs, such as peristalsis in the digestive tract or blood pressure regulation. The latch state mechanism further enhances this by allowing myosin to remain attached to actin even as dephosphorylation occurs, slowing their detachment rate. These features ensure that smooth muscle can maintain contractions for extended periods with minimal energy expenditure, a key advantage for organs that need to function continuously without fatigue.
