Your body has its own internal cannabis system that regulates everything from mood to appetite.
In short: The endocannabinoid system is a vast biological network present in animals as simple as hydras. It uses internal neurotransmitters to bind with receptors throughout your brain and body, acting as a master regulator for memory, pain, immune function, and even the timing of pregnancy. It is your body's hidden homeostatic control.
The endocannabinoid system (ECS) functions as a complex signaling network composed of endocannabinoids, their receptors, and the enzymes that synthesize or degrade them. The two primary receptors, CB1 and CB2, were identified in 1990 and 1993, respectively. While CB1 is predominantly found in the brain and nervous system, CB2 is more common in the immune system. These receptors act as molecular targets for endocannabinoids like anandamide and 2-arachidonoylglycerol (2-AG), which are produced on-demand to modulate neuronal signaling.
A core mechanism of the ECS is retrograde transmission, where a postsynaptic neuron releases endocannabinoids that travel backward to bind with receptors on the presynaptic terminal. This process, known as presynaptic inhibition, allows the system to fine-tune neural activity by reducing the release of neurotransmitters. This mechanism is essential for synaptic plasticity, including long-term depression, and helps the brain selectively inhibit excitability over varying time scales. Beyond the brain, the ECS influences peripheral tissues, including skeletal muscle, where it plays a role in insulin sensitivity, mitochondrial metabolism, and protein homeostasis.
The system is highly responsive to stress and environmental cues. For instance, the ECS helps habituate the hypothalamic-pituitary-adrenal axis to repeated stress, with anandamide and 2-AG playing divergent roles in regulating corticosterone responses. Research also suggests the ECS is involved in food-seeking behavior, with hypothalamic endocannabinoids acting as a tonic regulator of hunger. Because of its wide-reaching influence on physiological processes—from motor coordination to the modulation of orofacial musculoskeletal pain—the ECS is a major target for potential therapeutic interventions in conditions ranging from obesity and diabetes to chronic pain and muscle dystrophies.
